Other than the Free Bus, single bus tickets cost £2 - you can tap your debit/credit card. You can get a Day Rider for £5, which works on all busses, all day.

From Piccadilly Train Station:

Get the Free Bus (#2) from Piccadilly Train Station to Cheapside (Stop CL); then walk 2 minutes to St. Ann's Church (click for map)

Get on the bus outside the front of Piccadilly station (exit by "Boots"); there is a large circle with a number of bus stops. Free Bus #2 goes every 10 minutes.

From Victoria Train Station:

Walk 11 minutes to St. Ann's Church (click for map), or;

Get the 67/8/100 bus from outside Victoria Train Station (Stop NY) to Barton Arcade (Stop NA); then walk 2 minutes to St. Ann's Church (click for map)
